Background

Fixable Viability Dye 452 is excited by the Violet laser (with an excitation maximum of 405 nm) and has fluorescence emission maximum at 452 nm, Fixable Viability Dye 452 has similar emission to Brilliant Violet 421™.

Fixable Viability Dye 452 can intracellular diffusion of the dye and covalent binding to higher overall concentrations of amines, Each lot of this product is quality control tested, is useful for discrimination of viable from non-viable mammalian cells in multicolor flow cytometric applications.

Each lot of this product is quality control tested. The labeled cells can be fixed with formaldehyde for downstream decontamination, freezing and/or permeabilization and subsequent intracellular staining while maintaining stable viability stain fluorescence.

Protocol

1.Wash cells one time in PBS buffer.

2.Add 1 µl of Fixable Viability Dye 452 Stock Solution for each 0.5~1 ml of cell suspension/PBS (1:500~1:1000) and vortex immediately. It is recommended that the reagent be titrated for optimal performance for each application.

3.Resuspend cells at 1*10^7 cells/ml in Fixable Viability Dye 452, Incubate the mixture for 10-15 minutes at room temperature protected from light.

4.Wash cells twice with PBS buffer or equivalent buffer containing serum or BSA.

5.Resuspend the cells in Stain Buffer.

Note: This protocol applicable to Human PBMCs (human peripheral blood mononuclear cells) Dying, the reagent be titrated for optimal performance for different cell types. Stain, fix and permeabilize cells as desired for downstream applications.

Wow, I don't even know what to say about this book, but well done. I was a little bit mystified in the beginning. Our FMC and MMC definitely had me feeling a specific sort of way. A father, who was released from prison after "The Event" where all the white people in the Nation killed themselves, struggles with his identity. His daughter, Sidney, a biracial young adult who was raised by her white family, struggles with the fact that things she was taught were not necessarily true, and finds it hard to connect to her blackness. This is a tumultuous story of a beautifully written adaptation to an entirely new way of life. I didn't necessarily agree with certain ideations of the author, who in his book, claimed that there was a lost feeling after "The Event," but that's just personal experience, and I think that many black people would adapt. This book brings up so many feelings. How would I feel if "The Event" happened? Now that the "system" is dismantled? This book brings up dialogue and conversation. Amazingly well done. I'm still pondering. I know that this will be something I think about often. Wow. Very seldom do I tear up at books, but the representation of black joy in this story genuinely made my heart feel full. The love that when black people come together with nothing but amazing energy, was described so adequately in this book. I could feel it myself. And to that, I have to applaud Cebo Campbell.
