Rapport-Vassiliadis R10 Broth is used for the selective enrichment of Salmonella spp. from meat products, feces, and water polluted by sewage. The medium was formulated by Rapport and modified by Vassiliadis by reducing the concentrations of Malachite Green and Magnesium Chloride. Enzymatic Digest of Casein provides amino acids, peptides, carbohydrates, vitamins and electrolytes for bacterial growth. Sodium and Magnesium Chloride supply essential electrolytes and maintain the osmotic equilibrium. Potassium Dihydrogen Phosphate serves as a pH buffer. Malachite Green specifically enriches for Salmonella, since other species are typically resistant to or inhibited by this compound. There is a choice of 3 sizes - 500g, 2kg, and 10kg.
PREPARATION: Mix 26.6 grams of the medium in one liter of purified water until evenly dispersed. Heat gently with repeated stirring to dissolve completely. Autoclave at 116°C (10 lb. pressure) for 15 minutes.
