Background
Disabled-2 (DAB2) is a multifunctional adaptor protein involved in various cellular processes, including clathrin-mediated endocytosis, signal transduction, and regulation of key signaling pathways such as Wnt, MAPK, and TGF-β. Initially identified as a tumor suppressor in ovarian cancer, DAB2 is downregulated in several cancers, including breast, lung, and colorectal cancer, contributing to tumor progression. It contains functional domains like the phosphotyrosine-binding (PTB) domain and proline-rich SH3 domain, which enable it to interact with multiple proteins and regulate pathways involved in cell differentiation, proliferation, and epithelial-mesenchymal transition. DAB2 also plays a role in platelet activation and aggregation, with different isoforms expressed in human and mouse platelets. Additionally, it is essential for embryonic development and maintaining epithelial cell polarity.
