Background
TACSTD2 (Trophoblast Cell Surface Antigen 2, also known as TROP2 or EGP1) is a type I transmembrane glycoprotein belonging to the GA733 gene family, originally identified on the surface of human placental trophoblast cells. Functionally, it serves as a calcium signal transducer that regulates cell proliferation, differentiation, and apoptosis through downstream pathways including MAPK and PI3K/AKT, while also participating in embryonic development and tumorigenesis. Clinically, TACSTD2 is highly expressed in various solid tumors, including triple-negative breast cancer, urothelial carcinoma, and lung cancer, and is associated with tumor invasion, metastasis, and poor prognosis. The antibody-drug conjugate Sacituzumab Govitecan (SG), developed against this target, delivers the topoisomerase I inhibitor SN-38 specifically to tumor cells and has demonstrated significant survival benefits in multiple phase III clinical trials for metastatic triple-negative breast cancer, HR+/HER2- breast cancer, and urothelial carcinoma, representing a major breakthrough in pan-cancer therapeutics.
