Background

SLC6A1 encodes GAT-1, the principal Na⁺-coupled transporter that clears the inhibitory neurotransmitter GABA from the synaptic cleft by pumping it back into neurons and astrocytes, thereby terminating GABAergic signaling and maintaining excitatory–inhibitory balance throughout the CNS; mutations that misfold GAT-1 or abolish its plasma-membrane trafficking reduce synaptic GABA clearance, causing epilepsy, intellectual disability, autism spectrum disorders, ADHD, and mood dysfunction, whereas chemical or pharmacological chaperones can rescue some variants by restoring correct folding and surface expression.
