Background
Phospho-PDGF Receptor α (Tyr762) refers to the activated form of the platelet-derived growth factor receptor alpha subunit that has been phosphorylated at its tyrosine residue at position 762. Following PDGF ligand binding and receptor dimerization, autophosphorylation occurs, and the Tyr762 site serves as a critical docking site. Its primary function is to specifically recruit and bind downstream signaling proteins containing SH2 domains, such as Growth Factor Receptor Bound Protein 2 (Grb2), thereby activating the Ras/MAPK signaling pathway. This phosphorylation event is a central step for the receptor to transmit extracellular mitogenic signals into the nucleus, directly regulating key biological processes like cell proliferation, survival, migration, and differentiation. Consequently, detecting the phosphorylation level of the Tyr762 site is an effective method for evaluating the activation status of PDGFRα and the activity of its downstream signaling pathways, holding significant importance as a biomarker in research areas such as cancer, fibrosis, and targeted drug development.
